Reconstituting lyophilised peptides

Research peptides ship as a lyophilised (freeze-dried) powder because the dry solid is more stable to store and transport than a solution. Before use in an assay the powder is reconstituted — dissolved in a diluent to a known working concentration. This is a general laboratory method; always defer to the compound’s Certificate of Analysis and your own protocol.

Choosing a diluent

Sterile water and bacteriostatic water are common laboratory diluents. Solubility varies by compound — some require a specific solvent noted on the COA. Add the diluent volume that gives the concentration your protocol calls for.

Calculating a working concentration

Concentration is mass divided by volume. For example, reconstituting a 10 mg vial in 2 mL of diluent gives 5 mg/mL; in 1 mL it gives 10 mg/mL. Choose the volume that lands on a convenient stock concentration for your assay.

Technique

Add the diluent slowly down the inside wall of the vial rather than directly onto the powder. Swirl gently to dissolve; do not shake, which can denature or foam the peptide. Allow it to go fully into solution and inspect for clarity before use.

Storing the reconstituted solution

Aliquot to avoid repeated freeze-thaw cycles, and store cold per the compound’s stated conditions. See storing research peptides and the per-lot values on its Certificate of Analysis.
